On February 29, 2008, only minutes after free agency started, he reportedly had a meeting scheduled with the Philadelphia Eagles. Samuel agreed to a six-year, $56 million contract with the Eagles later in the day. He was quoted as saying, “I just want to be able to win and get back to the Super Bowl,” shortly after signing. On August 22, 2008, during the third week of the preseason, he then returned to New England.

On September 7, 2008, Samuel played his first regular-season game for the Eagles against the St. Louis Rams. Due to his outstanding performance, which included 4 interceptions and 1 touchdown, he was later named a Pro Bowl Reserve. His first with the Philadelphia Eagles and his second Pro Bowl appearance. He set an NFL record on January 4, 2009, when he returned an interception for a 44-yard touchdown. This was his fourth postseason interception return for a score. He tied Don Burroughs (1960) and Ed “Bibbes” Bawel (2009) for the second-most interceptions in a season in Eagles history with his nine picks in 2009. (1955). In acknowledgment of his outstanding season, he was then selected to play in the 2010 Pro Bowl as a starter. He was chosen for the 2010 Pro Bowl for the fourth time.
